I believe it is a reference to being an aide, or assistant, to a bootlegger; a la ‘Girl Friday’. For many, being a bootlegger at that time wasn’t reputable at all, and being a bootlegger’s Friday would be just as disreputable.
Someone explained it a few days ago. Think Robinson Crusoe’s man, Friday. At the time a bootlegger was a low-life criminal and so to be a Bootlegger’s Friday was a pretty low insult. Wish I could remember what day it was said so I could credit the person with it, I’m just repeating it.

Bootlegger’s Friday baffles me, I know this strip was done during prohibition, but can’t make the connection.
